Effect of Durabolin, Cortisone and L-phenylalanine on Chloramphenicol Toxicity


Abstract
The hematological effects of anabolic steroid (durabolin), adrenocorticosteroid (cortisone) a-~nd L-phenylalanine of albino rats on chloramphenicol toxicity was studied vrith the follo;ving results:
1. Control rats receiving 0.15m1. of saline daily by intramuscular injection for four weeks, exhibited no significant changes in hemoglobin, reticulocyte, neutrophil as well as platelet before and after experiments.
2. In rats administered chloramphenicol intramuscularly at 50mg/kg of body -eight daily for four weeks, decrease of hemoglobin, reticulocyte, neutrophil and platelet accompanied by both decreased marrow nucleated cell count and megakaryocyte count and a markedly prolonged plasma 69Fe disappearance half time were noted.
3. In rats given a concomitant intramuscular injection of chloramphenicol (50mg/kg/day) and durabolin (25mg/kg/week), hemoglobin was less decreased as compared with that of tke only chloramphenicol-administered group. However, both reticulocyte and neutrophil were rather in-creased, whereas platelet was not changed before and after experiments.
Compared with those of the chloramphenicoladministered group, marrow nucleated cell count, megakaryocyte count, and RBC 69Fe uptake tended to increase and on the other hand, plasma 6¢¥Fe disappearance half time was significantly shortened.
4. In rats administered a concomitant daily i~itramuscular injection of chloramphenicol (50 mg/kg) and cortisone (0.8mg/kg) , both hemoglobin and reticulocyte were increased but neutr ophil and platelet were not altered significantly before and after experiments.
A trend of increased marrow nucleated cell aunt as well as increased RBC 69Fe uptake with a significantly shortened plasma 69Fe disappearance half time was found.
5.. In rats treated with chloramphenicol (50mg/ kg/day, I.M.) and L-phenylalanine (100mg/kg /~iay, P. 0.) for four weeks, hemoglobin and reticulocyte were significantly increased althouglh both neutrophil and platelet remained unchan;ed before and after experiments.
In addition, the marrow nucleated cell count, megakaryocyte count and RBC 69Fe uptake showed no appreciable difference from those of tl:ie chloramphenicol-administered group.
Only the plasma 69Fe disappearance half time was significantly shortened.
6. Among the three therapeutic agents, cortisone and L-phenylalanine disclosed the most excellent results and durabolin the least.
